Technical expertise includes: >Plasmid DNA construction and engineering, AAV capsid vector engineering. >Protein expression using bacterium, yeast and mammalian systems, and protein purification using a variety of techniques including AAV vector production and purification. >Development, purification and characterization of monoclonal antibodies; successfully isolated variable domains of heavy chain and light chains, construction of scFvs and F(ab)2 and Fabs, humanization of mouse monoclonal antibodies. >Biochemical assay development including ELISA, Enzymatic functional assays. protein phosphorylation and glycosylation verification, as well as protein KD measurement using Octet, SPR instruments. >Culturing mammalian cell lines and primary from different origin and development of a variety of cell-based assays, including AAV transduction, cell proliferation assays, cytokine secretion assays, response element-reporter assays using luminescence, flourescence and quanti-blue as read-out. >Scientific data preparation and analysis using the following software: Vector NTI, DNASTAR, BioEdit, Snapgene, Geneious, Graphpad Prism, Softmax, PyMOL et al.
